Comparison of gene expression changes in two wheat varieties with different phenotype to strip rust using RNA-Seq analysis

چکیده

The fungus Puccinia striiformis f. sp. tritici (Pst) is an important threat to wheat production because it can cause stripe rust. present study aimed to identify new rust resistance genes and provide a theoretical practical basis for breeding varieties with broad spectrum, stable, durable resistance. Wheat leaves inoculated Chinese yellow 34 were collected at different time points transcriptomic analysis based on the susceptible AVOCET S (AVS) AVSYr15NIL [near-isogenic line (NIL) derived from AVS]. results showed that the number of upregulated in two was 294, 364, 398, 604, downregulated 520, 178, 570, 345 1st, 3rd, 5th, 7th days post inoculation, respectively. Gene Ontology Kyoto Encyclopedia Genomes enrichment analyses found differentially expressed peroxisome proliferators-activated receptor signaling pathways, plant–pathogen interaction, styrene acrylic acid biosynthesis encoded protein kinases, signal transduction, transcription factors, functional components. Differentially randomly selected quantitative reverse PCR analysis, change trend same as in transcriptome data. this suggest related response could be valuable understanding mechanisms involved

Whole transcriptome expression analysis and comparison of two different strains of Plasmodium falciparum using RNA-Seq

The emergence and distribution of drug resistance in malaria are serious public health concerns in tropical and subtropical regions of the world. However, the molecular mechanism of drug resistance remains unclear. In the present study, we performed a high-throughput RNA-Seq to identify and characterize the differentially expressed genes between the chloroquine (CQ) sensitive (3D7) and resistan...

Diversity of Resistance Gene Analogues in Rust Resistance and Susceptible Bread Wheat Varieties

Fungal diseases, especially leaf and stripe rusts are wheat yield reducing factors in Iran and the world. In this study, genetic diversity of 20 wheat varieties with different response to leaf and stripe rusts were studied using primers designed based on the conserved regions of plant disease resistance genes. The banding patterns of polymorphic markers were scored as dominant and number of amp...

RNA-Seq Data Comparison with Gene Expression Microarrays

RNA-Seq is a powerful sequencing-based method that enables researchers to discover, profile, and quantify RNA transcripts across the entire transcriptome. Because the method does not require probes or primers, the generated data are completely unbiased, allowing for hypothesis-free experimental design. The ability to perform this type of analysis provides researchers a powerful tool for transcr...

Comprehensive Computational Analysis of Protein Phenotype Changes Due to Plausible Deleterious Variants of Human SPTLC1 Gene

Genetic variations found in the coding and non-coding regions of a gene are known to influence the structure as well as the function of proteins. Serine palmitoyltransferase long chain subunit 1 a member of α-oxoamine synthase family is encoded by SPTLC1 gene which is a subunit of enzyme serine palmitoyltransferase (SPT). Mutations in SPTLC1 have been associated with hereditary sensory and auto...
